Zithromax (Azithromycin) Tri-Pak (500 mg Tablets)

The Zithromax TRI-PAK (500 mg qd for 3 days) is indicated for acute bacterial exacerbations of COPD (chronic bronchitis) due to Haemophilus influenzae, Moraxella catarrhalis, or Streptococcus pneumoniae.

The Zithromax TRI-PAK offers a combination of benefits in the treatment of acute bacterial exacerbations of chronic bronchitis. In a large, randomized clinical trial, a 3-day regimen of Zithromax 500 mg delivered clinical success rates (85%, n= 125/147) comparable with a 10-day course of Biaxin® (clarithromycin) (82%, n= 129/157). Additionally, in 11 published European studies, a 3-day regimen of Zithromax demonstrated proven clinical success in the treatment of AECB.

Zithromax is rapidly and extensively taken up by leukocytes, which deliver the drug to the site of infection, where Zithromax maintains high sustained concentrations. These sustained tissue concentrations of a 3-day regimen of Zithromax allow for a short course of therapy.*

The Zithromax TRI-PAK also offers excellent tolerability with a low incidence of adverse events, similar to that of the Z-PAK®.In a large, randomized trial, the most common side effects of a 3-day regimen of Zithromax and a 10-day regimen of Biaxin were diarrhea, nausea, and abdominal pain, with comparable incidence rates for each symptom of 5% to 9% between the 2 treatment arms. In this study, no patients treated with a 3-day regimen of Zithromax discontinued therapy due to treatment-related adverse events compared with 2.5% of those with a 10-day course of Biaxin.

Zithromax has not demonstrated an inhibitory effect on the cytochrome P450 system. Zithromax is designated pregnancy category B.

Zithromax is contraindicated in patients with known hypersensitivity to any macrolide antibiotic. If an allergic reaction occurs, the drug should be discontinued and appropriate therapy instituted. Physicians should be aware that reappearance of the allergic symptoms may occur when symptomatic therapy for the allergic reaction is discontinued.

Pseudomembranous colitis has been reported with nearly all antibacterial agents. It is important to consider this diagnosis in patients who present with diarrhea.

The most common side effects of Zithromax TRI-PAK are diarrhea/loose stools (4%-5%), nausea (3%), and abdominal pain (2%-3%).
